BIOSAFETY LABORATORY LEVELS

Biosafety Level 1 Signage:

The signage may contain the name of the agent(s) in use and the name and phone number of the laboratory supervisor and/or other responsible personnel.

Biosafety Level 1:

Applicable for work that involves well-characterized agents that are not necessarily known to cause disease in immunocompetent adults, and present minimal hazard to the laboratory environment and personnel nearby. All bacterial, fungal, parasitic, viral, rickettswial, and chlamydial agents that have been assessed for risk but do not belong to a higher risk group can be safely handled at a BSL-1.

Biosafety Level 2 Signage:

Information on the signage must include the name of the agent(s), biosafety level of the laboratory, supervisor’s name and/or other responsible personnel, contact number(s), and mandatory procedures for entering and exiting the laboratory facility.

Biosafety Level 2:

Involves agents that present moderate hazards to personnel and the environment. Access to the laboratory is restricted when work is being conducted and laboratory personnel are trained to handle pathogenic agents and are supervised by scientists that are competent in handling infectious agents and associated procedures. All procedures involving infectious aerosols or splashes are conducted in BSC’s or other physical containment equipment.

Biosafety Level 3 Signage:

Signage must contain the name of the agent(s) that are being stored or handled, the biosafety lab’s safety level, supervisor’s name and/or responsible personnel, contact number, and mandatory procedures for entering and exiting the biosafety laboratory.

Biosafety Level 3:

Suitable for clinical, diagnostic, teaching, research, or production facilities that involves agents that can cause serious or potentially lethal disease through the inhalation route when exposed to. BSL 3 laboratories have significant engineering and construction features. Must utilize special safety practices, equipment and facility requirements. Potentially infectious materials must be placed in a durable, leak proof container during collection, handling, processing, storage, or transport within a facility.

Laboratory doors must be self-closing and should contain locks in accordance with the institutional requirements. The laboratory must be segregated from areas that are open to unrestricted traffic flow within the building. Laboratory access is restricted. A change room may be included in the passageway between the two self-closing doors to facilitate appropriate dress codes.

A hands-free or automatically operated sink must be implemented into the laboratory, for increased safety measures. The sink should be located near the exit door. A sink must also be available for hand washing in each unit if the biosafety lab is segregated into different compartments. Additional sinks may be required when risk assessment is completed.

The laboratory must be constructed in order to be cleaned and decontaminated with ease. Carpets and/or rugs are not permitted. Seams, floors, walls, and ceiling surfaces must be completely sealed. Spaces or cracks that are located around door frames, as well as ventilation openings should be capable of being sealed to promote space decontamination. All floors must be slip resistant, impenetrable to liquids, and chemically resistant in the event of a chemical spill or leak. Laboratory furniture and accessories must be accessible for cleaning and disinfecting. Bench tops and desk must be impervious to liquid and resistant to heat, organic solvents, acids, alkalis, and other hazardous materials.

Vacuum lines must be secured with HEPA filters, or a comparable filter.

HEPA filtration must be implemented for clean air that is discharged into the laboratory. The HEPA filters should be tested and/or changed at least annually.

BIOSAFETY LEVEL 4:

This level works with dangerous and exotic agents that present a high individual risk of aerosol-transmitted laboratory infection and life-threatening disease that is frequently fatal, for which there are not vaccines or treatments, or related agency with unknown risk of transmission. Level 4 is known to work with agents such as smallpox that are extremely dangerous to the public. Every individual entering the laboratory must be advised of the likely hazards and meet specific entry requirements practiced by institutional policies. Only authorized individuals who are required for scientific or operational purposes are authorized to enter.

The facility and/or laboratory should contain a strong, secure locked door along with a agenda, or another system for cataloging the date and time of all individuals that enter and leave the laboratory. Authorized personnel’s clothing must be removed in the clothing change room. All persons that enter the laboratory must use appropriate laboratory attire. All persons leaving the laboratory must take a personal body shower. Used laboratory clothing must not be removed from the inner change room through the personal shower. The used laboratory clothing should be treated as contaminated materials.

According to the U.S. Department of Health and Human Services, the Class III cabinet must contain a pass-through dunk tank, fumigation chamber, or a comparable decontamination system for substances and equipment that fail to be decontaminated in the autoclave can be properly removed from the cabinet.

Class III cabinets should contain a HEPA filter on the supply intake as well as 2 HEPA filters on the exhaust outlet of the unit
